First off, when it comes to video, look at your GPU and your video memory. Unless you have a laptop with shared memory, RAM and video RAM are not the same.
If you are playing videos off a DVD-ROM or CD-ROM drive, your fragmentation and RAM will not have anything to do with it. It could be the speed of your DVD-ROM or CD-ROM.
If you are playing the videos off the hard drive, my guess is that it would be video codec issues or your video card is not capable of handling the video. Are they high compression, high fidelity videos?
Just so you know, I would suggest defragmenting your computer once a week. I work with videos and graphics and do it every night. The more you use the hard drive the more you should defragment it.
